A casino is a facility where people gamble. They are seen throughout the Grand Theft Auto series; however, gambling and interactive casinos were not introduced until Grand Theft Auto: San Andreas.

All casinos are located in Las Venturas. These are:
- Caligula's Palace (accessible)
- The Camel's Toe
- The Clown's Pocket
- Casino Floor (accessible)
- Come-A-Lot
- The Emerald Isle
- The Four Dragons Casino (accessible)
- The High Roller
- The Pink Swan
- Pirates in Men's Pants

There are three accessible casinos where Carl Johnson can gamble. There is a variety of activities to wager money on. While playing a casino game, Carl can borrow money from the casino, leading to a negative money value. If the money is not repaid, he will get phone calls from loan sharks threatening him to pay up soon or he will be physically harmed. If money is still not repaid, loan sharks armed with Micro Uzis and AK-47s will show up in black Vincents and attempt to kill him. The loan sharks carry a lot of money, so the debt can be repaid by simply killing them and picking up the cash they drop. If Carl travels outside of Las Venturas, loan sharks will cease to spawn.
Inside all accessible casinos, three songs that feature on Master Sounds 98.3 can be heard playing:
- Maceo & The Macks' "Cross The Tracks (We Better Go Back)"
- Bobby Byrd's "Hot Pants - I'm Coming, I'm Coming, I'm Coming"
- James Brown's "Funky President"
All accessible casinos in the game are protected by security guards, so any unruly behavior will cause them to react aggressively and Carl will be unable to use any of the casino's facilities until he exits the casino and re-enters it. The guards are also programmed to behave like police officers, unlike strip club and regular guards, as Carl will gain a one-star wanted level if he fires a gun inside the casino or aims a weapon at them, while two stars are gained if he kills a guard with any weapon.
